QTL mapping and expression analysis of candidate genes for root traits in riceChinese Full Text

Abstract: Recombination Inbred lines(RILs) were bred from an indica rice cultivar Huazhan(HZ) as the male parent and a japonica rice cultivar Nekken2(Nekken2) as the female parent. Root traits of the recombinant inbred lines at the trileaf stage were measured by scanning. Based on a high-density genetic map distributed evenly on 12 chromosomes, QTLs for rice root traits were mapped and analyzed. The results showed that three QTLs related to total root length were detected on chromosome 2, 5 and 10, respectively. Three QTLs related to root surface area were located on chromosome 2, 4 and 5 of rice, respectively. Six QTLs associated with mean root diameter were located on chromosome 4, 5, 9, 10 and 11, respectively. QTL for total root length and root surface area overlapped on chromosome 5. QTLs for root surface area and mean root diameter overlapped on chromosome 4. At the same time, candidate genes in the detected QTL interval were screened, summarized and analyzed, and analyzed by qRT-PCR. The total root length LOC_Os05 g40180 gene, root surface area LOC_Os11 g24240 gene, root average diameter LOC_Os04 g52030 gene were found to be new genes that affected root traits of rice. These results would have certain reference value for exploring the influence of root traits on cultivating ideal plant type and improving rice yield and quality.
